Recent studies suggest that monocytes/macrophages play a role in the pathogenesis of Behcet's disease (BD). The chemokine monocyte chemoattractant protein-1 (MCP-1) and its receptor, CCR2, are key regulators of monocyte trafficking. The aim of this study was to examine MCP-1 and CCR2 expression in BD. Serum samples and peripheral blood mononuclear cells (PBMCs) of 24 BD patients and 24 healthy controls were studied. Peripheral blood mononuclear cells of BD patients were isolated and stimulated with lipopolysaccharide (LPS) or interferon-gamma (IFN-gamma). Subsequently, MCP-1 mRNA expression was measured by real-time polymerase chain reaction. MCP-1 protein levels in supernatants of activated PBMCs were measured by ELISA. MCP-1 plasma levels were significantly higher in BD than in control subjects (P < 0.05). MCP-1 mRNA levels were higher in BD patients than in controls (P < 0.05). MCP-1 mRNA levels were higher in BD patients with gastrointestinal symptoms than in patients with skin lesions (P < 0.05). The peak MCP-1 mRNA levels in BD patients were observed at 5 hours of LPS stimulation. Activated PBMCs supernatants of BD patients revealed significantly higher levels of MCP-1 than control subjects (P < 0.05).
